In order to clarify the role of the cytokines in growth and differentiation of the mast cells, bone marrow cells from W/W^ν mice and its congenic +/+ mice were cultivated in the methylucellulose culture. The effects of recombinant mouse stem cell factor (SCF) and recombinant mouse interleukin 3 (IL-3) and interleukin 4 (IL-4) were examined. It has been shown that IL-3 alone stimulated growth of bone marrow cells from both W/W^ν mice and +/+ mice, and promoted the differentiation to the mast cells including a number of cytoplasmic granules which were stained with toluidine blue and Alcian blue but not with safranin, whereas neither IL-4 nor SCF stimulated individually. SCF, however, strikingly promoted growth of mast cells from +/+ mice, and induced maturation to the safranin-positive mast cells with the combination with IL-3 or IL-4 or both of them. The collaboration of the three cytokines was the most effective on growth and maturation of mast cells from ever W/W^ν mice as well as +/+ mice. The similar results were confirmed in the flow cytometry of the berberine stained mast cells growing under the influence of various combination of three kind of cytokines. It is suggested that mast cells should contact directly with fibroblasts to receive the proliferative stimulus, because SCF as c-kit ligands is expressed on the surface of fibroblasts. The cell-cell interaction between mast cells and Balb3T3 fibroblasts was observed in situ on the electron microscopy of co-culture with them. The ultrastractural views showed that the mast cells were highly activated in contact with the fibroblasts.
