To obtain osteoclasts, bovine bone particles were implanted intraperitoneally into low calcium-diet rabbits. The cells gathered around bone particles were induced to osteoclasts by culture. The most effective method to obtain precursors was, to rabbits breed on a low calcium-diet for 4 weeks, and then implant bone particles intraperitoneally for 3 weeks. When the cells harvested from the rabbits were cultured in a medium containing 10% serum obtained from a low calcium-diet rabbit, several TRAP-positive cells were observed after a 7-day culture. When normal-diet rabbit serum or fetal bovine serum was used, few TRAP-positive cells were observed. These findings suggest that the serum obtained from low calcium-diet rabbits contains some factors for differentiation of cells to TRAP-positive cells. The formation of resorption lacunae on the bone surface and the influence calcitonin suggested that the TRAP-positive cells were osteoclasts. In conclusion, the low calcium-diet rabbits used in this experiment were useful for harvest of precursors of osteoclasts, and the serum from low calcium-diet rabbits is effective to induce and differentiate osteoclasts in vitro.
