Porphyromonas gingivalisの株間遺伝子変異に関する分子系統学的研究

This study is aimed at clavifying the genomic diversity in 5 strains of Porphyromonas gingivalis (P. gingivalis) and 1 strain of Bacteroides forsythus (B. forsythus) by analyzing the nucleotide substitution of the whole length of an 16S rRNA, the elongation factor (EF)-Tu and EF-G and foc-like genes. Nucleotide sequences of 16S rRNA, EF Tu and EF-G genes in P. gingivalis were conservative in comparison with foc-like genes, although nucleotides were more or less mutated in all genes. In 16S rRNA genes, the region between the nucleotide positions 1003 and 1462 was found to contain many mutated positions within the strains, suggesting that this region might be useful for identification of a specific P. gingivalis strain level. The mode of nucleotide mutation in each gene was different when compared with others. Judging from the nucleotide sequences of 16S rRNA, EF-Tu or EF-G genes, the strains W83 and A7A1-28 were classified into a different phylogenetic group from the other strains FDC 381,SUNY 1021 and ATCC 33277. It is therefore concluded that genomic diversity among P. gingivalis strains is present even in nucleotide sequences of the mostly conservative 16S rRNA, EF-Tu and EF-G genes.
