An extracellular acid proteinase from C. albicans has been implicated in the pathogenesis of Candidosis. It has been shown that the proteinase breaks down many important proteins, such as immunoglobullins, hemoglobin, complement factors, albumin and collagen, which would implicate as a direct inflictor of damage on host cells as well as host defense system. Furthermore, the proteinase enhances vascular permeability by activating the Hageman factor-dependent pathway to release bradykinin, and may lead to inflammatory reactions in host. However, many questions of the role of a proteinase in the Candida infections are still remaining. In the present study, the role of the Candia proteinase in the growth, adhesion and systemic infection in mice by the yeast was examined using a specific aspartic proteinase inhibitor pepstatin. When C. albicans was cultured in a synthetic medium (yeast nitrogen base without amino acids and (NH_4)_2SO_4) containing bovine serum albumin as a nitrogen source, obvious growth of the cell was observed. However, the cell growth was strongly inhibited when pepstatin was added to the medium. The inhibitory effect of pepstatin was dependent on the concentration from 0.1 to 10 μg/ml. The antifungal effect of pepstatin was neither fungicidal nor fungistatic action. No effect of pepstatin was observed when the cells were grown in Sabouraud broth. Adherence of proteinase producing C. albicans strain to the cultured fibroblasts was inhibited by pepstatin. Pepstatin was protective when it was injected before the mice were infected with the yeast. However, little protective effect was observed when pepstatin was injected after the mice were received with the yeast The results suggest that an extracellular proteinase of C. albicans may play an important role in the growth of the cells, and that pepstatin has the possibility to be used as a new antifungal drug.
