The effects of unilateral malocclusion given experimentally on secretory functions of the submandibular glands (SMG) of SD rats which three left upper molar teeth were extracted and a left upper incisor tooth was amputated repeatedly at 3-day intervals were investigated. At 1,3,7,14 and 28 days after treatment, salivas elicited by i. p. octopamine (OCT), the selective α_1-adrenergic agonist, and isoproterenol (IPR), the selective β_1-adrenergic one, were collected and their volumes and proteins were compared among the extracted side of the experimental group (Ext), the non-extracted side of the experimental group (Non-Ext) and the control group. In addition, the various peptides were examined by HPLC and their amino acid sequences and homologies were determined and computed. The findings were as follows : 1. The wet weights of the SMG, expressed as mg/100 mg body weight, for Ext significantly increased by all days after treatment except at 14 days and for Non-Ext did at 1 day. In addition, for Ext they were significantly heavier than those for Non-Ext at 7 days. 2. The salivary volumes in response to OCT significantly increased at 1,3 and 7 days, whereas in response to IPR they were significantly reduced at 14 days after treatment. 3. The amounts of protein secreted in response to OCT significantly increased at 1 and 3 days, whereas in response to IPR they significantly did only at 1 day after treatment. 4. The esteroprotease activities of OCT-stimulated saliva significantly increased at 7 days after treatment. 5. The cystatin concentration of OCT- and IPR-stimulated salivas significantly increased at 7 days after treatment. 6. The salivary proteins in response to OCT were slightly changed quantitatively or qualitatively over a range of pI 3.5-4,whereas in response to IPR they were not changed greatly after treatment. 7. The optical density of 13 peaks eluted by HPLC was determined and compared among three groups. Many peaks for the Ext and Non-Ext groups significantly increased at 1 day, whereas the number of significantly different ones was reduced at 3 days. Over 7 days after treatment, with few exceptions, the peaks for Ext and Non-Ext were not significantly different from controls due to simultaneous increases of peak values with aging. But, in response to OCT at 7 days, and in response to IPR at 14 and 28 days, the 1 to 2 peaks for Ext were characteristically lower than those for Non-Ext. 8. Amino acid sequences of P20 (pentapeptide), P24 (dodecapeptide) and P31 (tetradecapeptide) were K-L-L-A-R, K-L-L-A-R-S-P-L-G-R-R-F and G-R-P-P-K-K-I-F-P-X-F-I-Y-R, respectively. These 3 peaks were presumed to be driven from two kinds of Glx-rich proteins, as seen in amino acid sequences. 9. The concentrations of the salivary peptides were markedly changed with aging and dependent on the nature of stimulus. It is concluded that the unilateral malocclusion given experimentally affects the secretory functions of the "bilateral" SMG of rats, and salivary peptides especially driven from Glx-rich proteins were drastically affected during this experiment.
