The effects of vitamin A overdose on the cellular dynamics of epithelia and the mesenchymal tissues of the secondary palate in embryonic rats were investigated. Ten-day pregnant rats received 2ml of vitamin A, 270μg/g body weight intraperitoneally for the induction of a cleft palate in rat fetus. The secondary palates of 15, 16, and 17-day-old fetuses were investigated by immunohistochemistry for Mcl-1, Bcl-xL, Bcl-2, Bak and Bax, and by TUNEL staining. New measurements were carried out for fetal growth dating. Fetal palatogenesis was the same as the previous reports. Secondary palates elongated dowonward to horizontally on fetal day 15, adhered on fetal day 16, and closed completely on fetal day 17 (when the epithelial seam disappeared). In the vitamin A-treated fetuses, proliferation activity of mesenchymal tissue necessary for palatal fusion decreased, and bilateral palates kept their horizontal position. It was suggested that cleft palate is induced by a poor combination of epithelial and mesenchymal growth. Epithelial apoptosis could not synchronize with mesenchymal growth at the secondary palate shelves, and early cellular differentiation might discourage proliferative activity of the mesenchymal tissue.
