Dental plaque formation is in part related to different functions of salivary glands and saliva properties. The present study was thus performed to elucidate whether there are differences in gene expression relating to secretory functions of submandibular glands (SMGs) between plaque-resistant (RES) and plaque-susceptible (SUS) rats. Steady-state mRNA expression levels of aquaporin (AQP) and adrenoceptor (AR) subtypes of SMGs were measured by RT-PCR and competitive RT-PCR. Unexpectedly, salivary flow rates and total protein secreted (μg/mg/h) in response to some stimulants by SUS rats were significantly faster or higher than those by RES rats. Four AQP (AQPs-1, -4, -5 and -8) and five AR (α_<1a>-, α_<1b>-, α_<2A>-, β_1- and β_2-ARs) subtypes were expressed in SMGs of RES and SUS rats. α_<1a>-mRNA was most abundant and more than 70% in total AR in SMGs of both groups at all ages. The expression intensities of four AQP subtype mRNAs in SUS rats were significantly higher than those in RES rats at all weeks of age with some exceptions. In contrast, mRNAs for AQP-8 in RES rats were more abundantly expressed than those in SUS rats at 48 weeks of age. For AR subtypes, α_<1b> at all ages, and β_1 at 48 weeks of age in SUS rats were significantly higher, whereas β_2 and the total AR at weeks of age were significantly lower in SUS rats. Relative rank orders of AR subtypes between RES and SUS rats at all ages were not greatly different. It is thus suggested that there are certain correlations among dental plaque formation, functional alterations for saliva secretion and mRNA expression of AQP and AR subtypes.
