The Effect of Gamma Irradiation on the Very Dilute Solution of Collagen in the Atmosphere of Air and Nitrogen

Low dosage gamma-ray irradiation on very dilute solutions of collagen are investigated. Acid-soluble collagen extracted from calf skin are fully purified and dissolved into a 0.05.% aqueous solution of acetic acid at a concentration of 0.021%. Gamma-ray irradiation with a dose rate of 4×10^2r/min are given to the solution in the atmosphere of air or nitrogen. Irradiation in air up to 3×10^4r produces about 70% decrease of intrinsic viscosity, about 20% decreases of optical rotation and of average molecular length estimated from flow birefringence, and noticeable lowering of denaturation temperature. The results suggest the occurrence of scission reactions in polypeptide chains in the triple helix of collagen molecule. On the other hand, irradiation in nitrogen up to 3×10^3r causes about 20% decrease of optical rotation, about 20% increase of average molecular length, and remarkable increase of intrinsic viscosity. Gelation takes place at about 5×10^3r. The results indicate the predominance of crosslink reactions during irradiation in nitrogen.
