Glutamate dehydrogenase (GDH) from leaves of 80 species of the Ranunculaceae were resolved by polyacrylamide gel electrophoresis into 10 anodal migrating isozymic bands, which were marked with 1-10 from the cathode toward the anode. They were charge isomers with a molecular weight of about 250,000. The GDH isozymes of each species showed multi-banded pattern, but usually one main band, or sometimes two, which was stained most intensively was distinguished. GDH of shoot apexes and that of roots showed different zymograms from that of leaves and from each other, but the position of the main band was not remarkably changed among these organs, except some species of Ranunculus and Coptis quinquefolia. The positions of the main bands in zymograms form cotyledons, hypocotyls including epicotyls and primary roots varied considerably, and often differed from those in zymograms from shoot apexes, roots and leaves. Nevertheless, most of main bands found in these seedling organs could be detected also in leaves though as minor bands. Although minor bands were variable in their position and staining intensity, the position of main bands in leaves was stable within a species and even within a genus. In Caltha, Trollius, Cimicifuga, Aconitum, Hepatica and Aquilegia, the band 6 was the main, and this is the commonest main band in the Ranunculaceae. In Clematis the band 5 was the main and in Thalictrum both the band 1 and the band 10 were the mains. In Anemone, Enemion, Dichocarpum and Coptis, the position of the main band varied within the genus but usually it is stable among the species belonging to the same subgenus or section. While in Ranunculus zymograms often differed with organs remarkably and the position of main band differed even among closely species. Ranunculus is an unstable genus about GDH isozymes.
