Physical mapping of 5S and 18S rDNA in lettuce, Lactuca sativa L.(Asteraceae)

Both 5S and 18S rRNA genes were physically mapped in the chromosomes of lettuce (Lactuca sativa L., Asteraceae) by the method of fluorescence in situ hybridization using homologous DNA probes generated by PCR. The 18S rDNA probes were hybridized with four chromosomal sites, which corresponded to the nucleolar organizing regions on the short arms of two satellite chromosome pairs, the third and fifth pairs in the order of length in the complement. The 5S rDNA probes were detected at the interstitial region of the short arm of one satellite chromosome pair, the fifth pair. No difference in karyotypes, banding patterns and localization of both 5S and 18S rDNA was noted in the seven cultivars of lettuce observed.
