Induction of mRNAs for Phosphoenolpyruvate Carboxylase and Pyruvate, Orthophosphate Dikinase in Leaves of a C_3 Plant Exposed to Light

We compared the genes for phosphoenolpyruvate carboxylase (PEPC) and pyruvate, orthophosphate dikinase (PPDK) from a C_3 plant, rice, with those from a C_4 plant, maize, using the maize cDNA for each enzyme. The cDNA inserts hybridized to several fragments of rice DNA digested with restriction enzymes, even under extremely stringent conditions, demonstrating the presence of multiple genes in rice with structures quite similar to those of the genes for PEPC and PPDK in maize. Northern blot analysis, using the cDNA inserts, revealed the presence of transcripts with sizes similar to those of the mRNAs for PEPC and PPDK from maize in the population of poly(A)^+ RNA from greening rice leaves, but not in that from etiolated leaves. The levels of the transcripts in fully green leaves were much lower than those in greening leaves, a result that corresponds to the changes in levels of proteins observed previously. The correspondence suggests that the expression of the transcripts was responsible for the synthesis of PEPC and PPDK proteins in greening rice leaves. These findings also indicate that the genes for PEPC and PPDK in rice not only have structures similar to those of the genes in maize, but are also regulated by light in a similar manner.
