1. The growth rate of cultured free-cells and their contents of chlorophyll, Mg, K and N reached peaks on media containing Σ M^<n+>=60 and 80 meq per liter, or in which π_<salts>=2.7 and 3.6 atm. These concentration levels in the media were considerably higher than those (π_<salts>=1.5-2.0 atm) which produced good growth of intact plants. This discrepancy in media concentrations may be due to differences in the culture conditions and in the morphology of the materials. 2. Very low Ca^<2+>/(Σ M^<n+>-Ca^<2+>) equivalent ratios of 0.1 to 0.01 in a medium were sufficient for good growth of free cells, while generally about unity of the ratio is required in a medium for good growth of intact plants. The difference in the medium composition may also be due to morphological differences in the materials. 3. Addition of Cl^- to the medium further promoted the growth rate and friability of cell clumps. 4. A low concentration of 0.01 mM BO_3^<3-> in the medium also promoted the friability of cell clumps.
