Protoplasts were isolated enzyatically from haploid tobacco leaves, then cultured in synthetic media. Cultured protoplasts started their first division within a few days after inoculation and subsequently developed into small colonies. After several reinoculations, these colonies differentiated shoots and roots. Finally, haploid flowering plants were obtained.
