Effect of abscisic acid on elongation and kinetin-induced tuberization of isolated stolons of Solanum tuberosum L.

The response of isolated stolons cultured in vitro, to abscisic acid (ABA) has been studied in the presence and absence of kinetin (6-furfurylaminopurine). ABA alone in concentrations from 7.5×10^<-4>mM to 7.5×10^<-2>mM, inhibited stolon elongation but failed to promote tuber initiation. In the presence of kinetin, ABA at concentrations of 3.0×10^<-2> and 7.5×10^<-2>mM markedly inhibited kinetin-induced tuber initiation and stolon elongation, but at 7.5×10^<-4> and 7.5×10^<-3>mM ABA did not prevent tuber initiation. When stolons were incubated on a medium containing kinetin and later transferred to one containing ABA with or without kinetin, the inhibitory effect of ABA decreased appreciably as the time of incubation on kinetin is increased. The results are discussed in relation to the role of ABA in the inhibition of nucleic acid and protein synthesis and the interaction with cytokinins and the possible effect of ABA on kinetin uptake, transport and accumulation at the locus of action.
