Preparation and Properties of Antisera against Lithocholic Acid Linked to Bovine Serum Albumin at the C-1β and C-6 Positions

• The preparation and antigenic properties of lithocholic aicd-bovine serum albumin conjugates in which the haptes are coupled to the carrier protein through a 6α-hemisuccinoyl, 6-O-carboxymethyloximino or 1β-hemisuccinoyl bridge are described. The antisera elcited by both the former conjugates showed relatively poor ability to discriminate lithocholic acid from ursodeoxycholic and chenodeoxycholic acids, whereas that from the latter revealed a high affinity constant (K_a=6.4×10^7M^<-1>) and excellent specificity to lithocholic acid, exhibiting no significant crossreactivity with any of the major human bile acids.
