The use of a chemically defined solution as the culture medium is of importonce in investigating planarian regeneration, since the results vary with differences in the ionic ratio. For example, the occuraence of supplementary eyes is affected by the ratio of sodium to potassium. Although it has been postulated by Child ('32) that diluted Ringer's solution is adequate for the cultivation of an asexual race of Dugesia gonocephala, the susceptibility of worms cultured in this solution to certain chemicals appered to be somewhat lower than that in well water. An attempt was therefore made to determine the compostion of an optimum solution for culturing the Japanese planarian, Dugesia gonocephala, based on the salt concentrations of the waters inhabited by the worms. Since it has been established that planarian worms can live well in media of which the ionic composition is rather simple, only the main cations were to be considered. The concentrations of sodium, potassium, calcium and magnesium in waters from four habitats of the worms were measured by flame photometry and by colorimetry. The hydrogen ion concentration of these waters was also determined. The results of the measurements are shown in Table 3. From these results the composition of the basic culture medium for Dugesia gonocephala, was found to be as follows : 6.4×10^<-4>M NaCl, 6.6×10^<-5>M KCl, 7.7×10^<-4>M CaCl_2, and 1.7×10^<-4>M NaHCO_3. The pH of the solution is near 7.4 It has since been observed that worms cultured in this solution are kept alive indefinitely.
