Wild type larvae of Drosophila melanogaster were treated with nitrogen mustard (methyl bis-β-chloroethyl-amine hydrochloride) or nitromin (methyl bis-β-chloroethyl-amine N-oxide hydrochloride). Larvae of 70 hours old were immersed in 1, 2 or 3% solution of these drugs and were kept for 1 to 3 hours in the dark room. Such treatment proved to be effective in producing anomalous flies. Anomalies were found in various organs of flies such as compound eye, leg, wing, abdomen, thorax and proboscis. Of these, compound eye was the organ showing the most frequent and the most marked anomalies (cf. Table 1). Anomalies of the eye were exclusively in the defective development of the facets. As a result, the compound eyes became Bar or eyeless like. In extreme cases, the facets produced counted as little as 20. There were other cases in which knob like protuberances equipped with bristles were found in the area of the compound eye which failed to produce facets. Anomalies of the leg showed mostly the deficiency in the distal part, especially in the tarsus. But sometimes duplicated legs were found to be produced. Most of the anomalous wings produced were notchy or Beadex like. But there were other cases in which black swellings were formed in the cell or in the vein. In the abdomen, fusion of the abdominal segments took place, and in the thorax and proboscis deficiency of scutum, scutellum and palpus occurred. Generally anomalies were produced more abundantly in the case of nitromin treatment than in the cases of nitrogen mustard. However, this does not necessarily imply that teratogenic effect of nitromin is stronger than that of nitrogen mustard. Because it was found at the same time that those larvae treated with nitrogen mustard failed to hatch more frequently than in the case of nitromin treatment. Presumably toxicity of the nitrogen mustard may be greater than that of nitromin.
