The chemical properties and the physiological effect of the nuchal gland substance upon the blood pressure of rabbits and toads were tested, using alcohol extract prepared after the method described in the previous paper ('53). Results obtained are as follows: 1. Height of the beat of isolated hearts increased 1.5-2 times the normal level by the addition of 0.1cc of 0.1% nuchal gland extract to the perfusion medium. This excitation continued for a few minutes. 2. Contraction of capillaries was observed both on the ear of the rabbit and on the hind limb of the toad when the extract was added to the perfusion medium and this contraction lasted about 1 hour or more. The effect of this type seems quite characteristic for the nuchal gland substance as compared with the transient one of adrenaline. After washing with fresh Ringer's solution, no recovery to the initial level was recognized. 3. Ninhydrin reaction was negative, indicating that the extract contains no protein or amino acid at a measurable amount. The ultraviolet absorption spectrum of the extract was very much alike to that of Epinephrine-HCl (adrenaline), but there was only one maximum at 210mμ in the former while the latter showed the 1st maximum at 220mμ and the 2nd one at 280mμ. Such a increase of blood pressure caused by the nuchal gland substance was attributed to the increase and maintenance of excitaion of both heart muscle and capillary wall.
