The perivitelline fluid was taken out of fertilized Oryzias eggs and its carbohydrate component analysed particularly by paper partition chromatography. The same technique was also applied to ana-lysis of a substance contained within the cortical alveoli. The perivitelline fluid was obtained by the following procedures. Unfertilized eggs were immersed in a 0.1 per cent CdCl_2-Ringer's solution for 10 minutes. The chorions of these eggs remain soft after fertilization and consequently the perivitelline space is enlarged considerably. Using a micropipette, the fluid contained was sucked out of the perivi-telline space of such eggs. It was found that the perivitelline fluid is a hydrophile colloid which contains polysaccharide. In another experiment the aqueous solution of the cortical alveoli was pre-pared, in order to obtain the alveolar material which has been known to be polysaccharide. Acid hydrolysates of these materials were analysed by ascending chromatography, for the identification of monosaccharides involved as components of the materials. Polysaccharide in the perivitelline space contains as monosaccharide components galactose, mannose and the sugar of presumably methylpentose, with the galactose predominating. The polysaccharide of alveolar content also shows the presence of the same monosaccharides, galactose, mannose and a sugar of presumably methylpentose with the galactose predominating as components. Judging from this mono-saccharide composition of the materials, it is highly possible to conclude that polysaccharide in the peri-vitelline space originates from the content of cortical alveoli by their breakdown on egg activation.
