The present observations have mainly been carried out to ascertain a correlation between the movement of the refractile bodies and the morphogenesis of the cyst membranes (an ectocyst and endocyst) at encystment in Stylonychia histrio S. Mull. The fact that all refractile bodies are densely packed in the cystoplasm from the precystic stage up to the encysting stage, shows that their periphery changes itself into the minute granules (α-granules), all of which are migrated from their central core part so as to be arranged beneath the pellicle. The result is that the central core is exposed. By the completion of the arrangement of the α-granules there is followed their disappearene, during which time an ectocyst, the outer cyst membrane, is formed by thickening of the pellicle. Thereafter, out of a part of the exposed refractile bodies in the encysted animals there is budded off the other kind of the minute granules (β-granules, ), which lie in the cavity between the ectocyst and the protoplasmic part, in which the β-granules similar to the α-granules disappear, when an endocyst, the inner cyst membrane, is completed. During the cystic stage, on the surface of the protoplasm there is the renewed pellicle, whose formation may take place by the change of the remaining refractile bodies which did not participated in the formation of the two cyst membraes. Any of the refractile bodies and of the various kinds of the above mentioned minute granules was of a sudanophilic and mitochondrial nature. In addition, though the central core of all the bodies has an affinity for the various bivalent metal cations during the vegetative stage, it is not proved whether the core has an affinity even after the precystic stage. It is supposed that the disappearence of an affinity has some relation to a state of protein contained in the core.
