The distribution of acid and alkaline phosphatases in prothoracic glands and corpora allata of the silkworm were histochemically investigated. The silkworms at different stages from 1st day of 4th instar to 2nd day after pupation were used as material, The freshly dissected prothoracic glands and corpora allata were placed on slide-glasses and were frozen by dipping them in a chilled methanol with dry ice, then they were desiccated in a freezing-drying-apparatus. These dried preparations were then incubated in the reaction medium. Acid and alkaline phosphatases were tested by the methods of Moog ('44) and Gomori ('41) respectively, using sodium glycerophosphate as substrate. 1) Prothoracic gland A mass of small particles was observed in nuclei on the 1st day of 4th and 5th instar (Fig. 1), and the activities of acid and alkaline phosphatases of these particles were found to be higher than those in other parts of the cells. On the 3th day of 4th instar, very small particles which showed high activities of both phosphatases, appeared near the surfaces of the cells. On the 5th day of 5th instar, lage granular figures rich in both the phosphatases began to appear near the surface of nuclei (Fig. 2). From observation on the preparation stained with haematoxylin and eosin (Figs. 4, 10), it seems that the granular figures mentioned above were identified with the parts of cytoplasm which were projected into nuclei. Approaching pupation, these granular flgures were decreased in number (Fig. 5). It was obsarved that the actiuties of both phosphataes were low at about 50 hours after pupation, and some of the cells were completely negative for both phosphatase reactions (fig. 6). 2) Corpora allata Numerous small particles stained with haematoxylin were present in nuclei through various atages, and these particles were found to have higher activities of both phosphatases than any other parts of the cells (Figs. 7, 8, 9). The difference of activities of both phosphatases between nucleus and cytoplasm was not observed distinctly. In the present observations on phosphatases of corpora allata, there were no remarkable changes in both phosphatases throughout molting and pupation, and activities of both phosphatases were maintained after pupation. In roder to observe precisely the activities and the distributions of phosphatases in the cells of prothoracic glands and corpora allata during larval development, these glands should be studies by section method. Thus, prothoracic glands at various stages were fixed with a chilled 80% alcohol solution or a achilled acetone, and were sectioned by the usual paraffin method. But it was found that by such procedures both the phosphatases were completely destroyed.
