Recently Karyological studies have been much advanced on the ascites sarcomas of white rats, but little has been known on the occurrence and nature of cytoplasmic granules in the tumor cells. The present paper describes the results of investigations on cytoplasmic granules in the tumor cells of the MTK-sarcoma I. The observations were carried out with the supravital staining method with neutral red or Janus green, and through smear method with Giemsa's stain. Azurophile granules: Smear preparations stained with Giemsa's solution demonstrate some azurophile granules which arrange in a characteristic rosette form. They were found distributed at one side of the kidney-shaped nucleus in the cytoplasm. Sometimes there occured some tumor cells in which the granules were evenly scattering in the cytoplasm. They were similar in size and round in shape. These azurophile granules ocurred most commonly in the tumor cells derived from the tumor-bearing rat on the 3rd to 7th day after transplantation of the tumor. Granules demonstrated by Janus green: The granules stained by Janus green were found scattered in the cytoplasm, assuming each a minute oblong shape. Granules demonstrated by neutral red: the application of supravital staining method with neutral red demonstrated the granules stained by this dye in the cytoplasm. As regard to the nature of the granules contained, the tumor cells of the MTK-sarcoma I were classified into two types as follows: I) R_1-type; the cells of this type contaian the neutral red granules which arrange in a rosette form in the cytoplasm. Sometimes, the granules can be seen colored light green. II) R_2-type; the cells belonging to this type show vacuoles stained by neutral red and varying in size. together with reddish or light green granules. Throughout a transplant generation of the MTK-sarcoma I, the daily frequency of the R_1- and R_2-type was studied. On the 3rd and 4th day after transplantation of the tumor, the cells of the R_1-type showed a high frequency in appearance, while those of the R_2-type were found abundantly on the 1st or 7th day. Since the actively dividing tumor cells were found most frequently on the 3rd and 4th day, it is supposed that the cells of the R_1-type are the active tumor cells, while those of the R_2-type are degenerating cells. The results from the experiments with α-peltatin solution served to supplement the above evidence. From the experiments carried out in this study, it was suggested tha the cytoplasmic granules demonstrated with Giemsa's solution, neutral red and Janus green seem to play an important role in the metabolic activity of the tumor cells.
