In order to account for the relationship between the propagation of spermatogonia and the functional activity of apical cells in the silkworm testis, histochemical investigations were undertaken, with special emphasis on polysaccharides and phosphatase. The results ob tained are summarized as follows. The apical cell f the silkworm testis seems to be derived from a part of the capsula lobuli, an internal membrane of the testicular follicle. It is probable that the spermatogonium and its cyst cell originate from the same primordial germ cell, i.e., after the division of a primordial germ cell, one of the daughter cells located on the opposith side of the apical cell is transformed into a cyst cell and envelops the other, thus the spermatogonium of one-cell stage is formed (Fig. 1). Moreover, it seems likely that the onset of spermatogonial formation has some relation to the matabolic activity of the apical cell. As spermatogonial multiplication continues from the end of the second instar to the beginning of the pupal period, a remarkable deposition of PAS positive granules occurs in the apical cell and a high concentration of phosphatase was shown in the spermatogonial region (Fig. 2, 3 and 6). From the end of the pupal period, however, the concentration gradually decreases becoming extremely weak when they emerge (Fig. 4, 7), at which time degeneration of spermatogonia begins (Fig. 8). These observations suggest that the apical cell functions as a nurse cell providing nutritive materials required for the development of young spermatogonia.
