The so called rodlet granules of Euplotes are always located near the bases of ciliary organelles. In conjugants, rodlet granules as well as surface organelles are newly made. In the present paper, the auther examines the process of the rodlet granule formation. At first a few tiny granules appear along the anterior edge of a peristomal rudiment, each being separated by two or three microns. In this case, none of granules has originated from pre-existing granules. The tiny granules increase rapidly in number, and soon form a single closely packed row. Some granules then move to the posterior edge of the peristomal rudiment, forming a second row of granules. Finally a third row of the granules appears between the first and second rows in the rudiment. The granules in each row increase in size, then differentiate into compound granule containing five to seven fine granules which stain deeply with acetic-methylgreen. They slowly lengthen, developing gradually into rodlets, and finally become arranged in rosette-fashion. When the new cirri are formed in the conjugants, the rodlet granules located near their bases are newly made too. In this case, the compound granules mentioned above are never observed. instead some of the new granules are constricted centrally and others seem to increase slightly in volume. these granules are scattered over the morphogenetic field of new cirri. Some of the granules then gather together near the base of each new cirri, and are arranged in rosette-fashion. None of these new granules originate from pre-existing granules.
