In females of the newt, Triturus pyrrhogaster, the cytoplasm of the oviducts shows a strong histochemical reaction of alkaline phosphatase during the breeding season. Even in the breeding season, however, newts kept at low temperature failed to produce a positive reaction of the enzyme in the oviducts (Table 1, I). If cooled animals were transferred from low temperature to room temperatuer, the enzyme activity of the cytoplasm of the epithelial cells of the oviducts was restored (Table 3, I). If the pituitary of cooled animals was removed at the time of transfer, the oviduct remained desitute of the enzyme (Table 3, II). It seems likely that a rise in temperatuer stimulates the pituitary of the newt to produce a substance which is effective in activating alkaline phosphatase in the oviducts. Ovariotomized newts with the pituitary intact exhibited a moderate enzyme reaction after transfer to room temperature (Table 3, III). Therefore, at least under the conditions of the present experiments the ovaries seem to play no important role in activating alkaline phosphatase in the oviducts. However, these results do not necessarily indicate that the alkaline phosphatase activity of the oviducts is totally independent of the ovaries (see Kambara, 1963a). The reaction of alkaline phosphatase in the epithelial cells of the oviducts was not attenuated within 7 days after ovariotomy, but by the 21st day it was nullified (Table 4, II). At three weeks after removal of the pituitary, all newts with their ovaries intact showed the enzyme activity in the oviducts (Table 4, III). A majority of newts six weeks after hypophysectomy still exhibited a positive reaction. PAS-positive substance in the cytoplasm of the epithelial cells of the oviducts was more readily affected by both ovariotomy and hypophysectomy. At 7 days after either operation, vacuoles unstainable with the PAS method occurred in the cytoplasm of the cells probably in association with degenerative changes in the cells (Fig. 1, B and E). It seems likely that in the cytoplasm of the epithelial cells of the oviducts the production of PAS-positive substance is more intimately related to the ovaries and the pituitary than is the activity of alkaline phosphatase.
