When split preparations of isolated caudal fins were soaked in Ringer's solution containing 5×10^<-7>M noradrenaline (NA), 5×10^<-7>M adrenaline (A) or 5×10^<-6>M dopamine for 5-6 min and then washed with Ringer's solution, the pigment-aggregating response of melanophores to KCl (K-aggregation) was augmented as compared with the response before the catecholamine treatment. The K-aggregation abolished by KCl+10^<-6>M reserpine could be restored, though temporarily, after soaking preparations in an A, NA or dopamine solution. Isoprenaline (5×10^<-5>M) was not effective for this restoration. Probably, the store of adrenergic transmitter which could be released by KClstimulation at the ending of the melanophore-aggregating nerve was increased by the uptake of NA, A or dopamine at the nerve ending, and the store depleted by reserpine was replenished with the exogenous catecholamine. The NA uptake by the nerve ending occurred even in a KCl+NA solution. Phenoxybenzamine inhibited K-aggregation. This inhibition remained after washing the preparations, but disappeared after the NA treatment. Both the restoration of K-aggregation in a reserpine-treated preparation and the augmentation by NA of K-aggregation in a normal preparation were inhibited by phenoxybenzamine. Pyribenzamine (antihistaminic) inhibited NA uptake by the nerve ending, but atropine did not.
