The distribution of RNA in spermatogonia, spermatocytes, spermatids and Sertoli cells at various stages of the cycle of the seminiferous epithelium in the mouse testis was observed in histological sections stained with methyl green-pyronin. RNA reaction in the nuclei was found only in the nucleoli of all the germ cells and the Sertoli cells. The nucleoli of the spermatocytes gradually enlarged during the pachytene and the diplotene stages, and then their central part appeared as pyronin-negative regions. RNA-containing materials in the cytoplasm were observed in all the germ cells and the Sertoli cells, especially in the type A spermatogonia and late spermatids in the course of spermiogenesis. The RNA materials of the spermatids increased their stainability in the caudal part of the cells through steps 9-13 (stages IX-XII and stage I) of spermiogenesis. And from step 14 (stage II) on, the materials were gradually integrated into larger granules, showing their maximum size and stainability at step 16 (stage VIII). The RNA materials in the residual bodies released into the lumina of the tubules with the development of the spermatids were observed at stages VIII and IX of the cycle of the seminiferous epithelium, and some of them were phagocytosed by the Sertoli cells during these two stages.
