The ultrastructures of the mitotic apparatus of synchronized L5178Y cells in Colcemid treatment and their changes in the recovery process from Colcemid were observed with electron microscope. The centrioles are not separated to the poles and are occasionaly surrounded by a group of chromosomes in Colcemid solution. Ten minutes after removal from Colcemid, many short microtubules are found radomly near the centrioles and kinetochores. Fifteen minutes after removal, the centrioles and chromosomes are connected with long microtubules. Chromosomes are arranged in a sphere which has the centrioles at the center. Mitosis proceeds in the following 30 minutes without typical arrangement of chromosomes in metaphase. This abnormal behaviour of the centrioles and the chromosomes is considered to be induced by the absence of microtubules in pro-metaphase caused by Colcemid-treatment.
