Sino-atrial region of carp hearts was excised and was pinned on a cork block. Transmembrane action potentials from spontaneously beating sino-atrial valve (S-A valve) were recorded by using the microelectrode technique. Slow depolarization, the pacemaker potential, during diastole was recorded from a large number of fibers in S-A valve. When the tip of the microelectrode was closely applied to the base of S-A valve, comparatively large amplitude of this potential was obtained. Cardiac nerve fibers have innervated into two leaves in sinus venosus. Effects of the cardiac nerve stimulation on spontaneous activity of S-A valve in the carp hearts were similar to those of vagal stimulation on that of sinus venosus in the frog heart and of sinus node in the mammalian hearts. When the nerve was stimulated relatively at a low frequency, the slope of the pacemaker potential was reduced, the resting potential was increased, and the rate of spontaneous beat was decreased. Repetitive stimulation at a higher frequency produced an arrest of a spontaneous beat, which may be caused by a suppression of the pacemaker potential and a hyperpolarization of the membrane potential. There was no significant change in the duration of the action potentials obtained during nerve stimulation. When stimulation ceased, the membrane potential depolarized again to the control value, and the spontaneous activity was resumed. The pacemaker fibers after stimulation showed an acceleration to levels above the control rate of spontaneous activity before reestablishment of a steady state.
