It has been reported that hematopoietic failure is responsible for the radiation death in fish as in mammals. A series of experiments, designed to clarify the cell population kinetics of hemopoiesis in irradiated and non-irradiated goldfish were carried out. Casassius auratus had been kept in water at 25, 15 or 6℃, and then irradiated with 1 or 8 kR of X-rays. ^3H-thymidine (10 or 15 μCi) was injected intraperitoneally into each fish just after irradiation. Fish were sacrificed at various intervals after the injection. Smear preparations of the head kidney (hematopoietic tissue) and the peripheral blood were fixed with methyl alcohol and coated with nuclear track emulsion of Sakura NR-M2 by dipping method for autoradiograph. In comparison of hematopoietic cell kinetics in fish kept at 25 and 15℃, the former was 3 to 5 times rapid than the latter. No marked inhibition of DNA synthesis was found immediately after lethal irradiation (8 kR) under the persent experimental conditions, whereas proliferation of hematopoietic cells was completely inhibited. Rate of disappearance of labeled cells from hematopoietic tissue was slower in cool environment. Following X-irradiation of 1 kR proliferation of the labeled cells delayed and cell death occurred. These results show that hematopoietic process in fish is observable by autoradiography with ^3H-thymidine and rate of development of radiation injury in the hematopoietic tissue is slowed down at lower temperature.
