Expression of estrogen receptor (ER) isoforms in the female reproductive tissues of neonatally diethyistilbestrol (DES) exposed mice

Perinatal estrogen treatment induces serious abnormalities in the reproductive organs. In this study, expression of ER α and ER β was examined using immunohistochemistry (IHC) in the female reproductive tissues of neonatally DES-exposed mice (DES-mice) to elucidate the roles of each ER isoform in the process. CD-1 mice were given daily s.c. injections of 4μg DES for 5 days from the day of birth (day 1 ) and killed at day 4 (3 injections), 10, 15 and 21 . Frozen sections were fixed with 4% paraformaldehyde and immunostained with anti-ER α (MC-20) or anti-ER β (Y-19); antibodies were obtained from Santa Cruz Biotech. Neonatal DES exposure did not influence the expression of ER β in the female reproductive tissues and of ER α in the ovary. ER β was localized only in the ovary. Immunoreaction of ER β in the follicular cells and ER α in the stromal cells were detected at day 4 and gradually increased with maturation. The DES treatment up-regulated the ER α expression in the epithelium and down-regulated it in the stroma in reproductive tract from day 4 to 15. At day 4, epithelial cells showed strong ER α-immunostaining in the oviduct, uterus, cervix and vagina in DES-mice; uterine epithelium was negative in vehicle-treated controls. The staining increased with maturation. These results suggested that ER α is the dominant ER isoform in the reproductive tract and participates to the irreversible changes in chracteristics of epithelial and stromal cells during the period of DES treatment through ER α system of both cell-types.
