The purpose of this study was to investigate the ultrastructural changes and functional changes of rat odontoblast by electron microscopy and Pb-EDTA labeling after vinblastine (VBL) administration. The main results were as follows : 1. Numerous Golgi-associated vesicles and coated vesicles of 50-80 nm and 90-110 nm in diameter accumulated in Golgi area of odontoblasts from 1 to 4 days after the intraperitoneal injection of VBL. 2. At this time, amorphous structures like fibrils were observed within Golgi saccules and black and white stripes were seen across filamentous structure. 3. Odontoblasts began to recover from 4 days and appeared normal 6 days after injection of VBL. 4. In the section of Pb-EDTA labeling, the rates of dentin formation were significantly inhibited during 2 days after injection of VBL. Decrease of lead sulfide was prominent in rat incisors. On the basis of the foregoing findings, VBL inhibited dentin formatiom and transport of Golgi-associated vesicles and coated vesicles, and pathway of collagen precursor includes the Golgi complex and entire microtubules system. On the other hand, there was no difference in the size and the number of microtubules 1, 2, 3, 4, 5, and 6 days after VBL administration, but the microtubules often appeared to wind in the odontoblasts except in the necrotic ones. Also, active secretion from mesenchymal cells like odontoblasts produced the structure like predentin under the necrotic odontoblast layer 6 days after injection of VBL.
