It is unknown when Langerhans cells (LCs) first enter the rat oral mucosa or acquire Ia antigen. In this study, development, distribution and density of LCs were studied in Wistar rat palatal mucosa from fetal day 16 to postnatal day 16 by using histochemical and immunohistochemical technique on epithelial sheets. LCs were defined as ATPase positive or Ia antigen positive cells. After birth, positively stained cells were counted and statistically evaluated. The results were as follows : 1. There were no ATPase positive LCs in the epithelium of palatal mucosa at fetal day 16. They first appeared at fetal day 18 and began to increase in number. Most of these cells were round or polygonal and only some of them were dendritic. But in the postnatal period dendritic cells became predominant. 2. There were no Ia antigen positive LCs in the epithelium of palatal mucosa at fetal day 20. They first appeared at postnatal day 0 and then these cells increased in number. Most of these cells were dendritic. 3. Daily changes in the density of ATPase positive cells were similar to those in the density of Ia antigen positive cells. But the density of ATPase positive cells was higher than that of Ia antigen positive cells throughout the ages examined. These results indicate that ATPase positive LCs matured into Ia antigen positive LCs after being exposed to some microenvironmental factors during perinatal period and then Ia positive LCs gradually increased by postnatal day 16.
