制癌剤軟膏の舌粘膜への影響に関する実験的研究

An experimental study was made with use of 75 ddY-mice of the effect of ointment type anticancer agents (BLM and 5-FU) on normal lingual mucosa. Furthermore, 5-FU concentration inside each tissue (tongue, liver and serum) was analyzed by GC-MF method. The results were as follows : 1. Piloerection, loss of color, debilitation of activity and decrease in body weight as systemic-macroscopic effects of topical application were observed throughout the experimental period. 2. These systemic-macroscopic effects were observed more serverely in the group of 5-FU than in the group of BLM. 3. Each ointment gave rise to the hyperactivity of epitherial cells (i.e. the body exchange of basal cells and prickle cells, increasing of mitosis) and increasing of keratohyalin granules. These findings were observed more in the group of BLM especially, being emphasized by the existence of wounds. 4. Increase keratohyalin granules prior to the keratinization of granule cells suggested its relation with the epitherial keratinization. 5. 5-FU concentration inside the tongue, liver and serum were beyond the useful concentration of humans (0.05μg/ml) ; the tongue especially showed a value three times higher, whereas the liver showed a value. As stated above, improvement in the means for retension of ointoment on the lingual mucosa is belived to make the use of this type of anticancer agents a useful chemotherapy for the oral and maxillofacial region, because drugs of this type are simple and direct in application, small in quantity required and less in side effect.
