T lymphocytes predominate in early periodontal lesions in human gingival tissue and such a lesion is called T cell lesion. In this study, an experimental T cell lesion was made by the injection of Concanavalin A (Con A) into rats' gingiva. The rats were sacrificed at 1st, 3rd, 7th and 14th day after the injection of Con A. Nucleated cells, lymphocytes, T cells and their subsets were counted in cell suspension from the gingival tissue and submandibular lymph nodes. Results were as follows ; 1. In gingival tissue, the nucleated cells, lymphocytes and T cells increased rapidly on 3rd day and decreased thereafter. 2. In submandibular lymph nodes, nucleated cells and lymphocytes increased gradually from 1st day to 7th day and decreased thereafter. 3. In submandibular lymph nodes, the number of T cells increased rapidly to a maximum level on the 3rd day. The results showed that T cells were affected by Con A in both the lymph nodes and the gingival tissues concurrently. 4. In submandibular lymph nodes and gingival tissue, the numbers of all T cell subsets (IgG_1, IgG_<2a> or IgG_<2b> positive cells) increased gradually after the injection of Con A. 5. In gingival tissue, the proportion of IgG_1 positive cells were greater than that of the other T cell subsets (IgG_<2a> or IgG_<2b> positive cells) in any days. 6. The profiles of changes in IgG_<2a> positive cell were similar to that in IgG_<2b> positive cell. 7. By the histological examination of gingival tissue, the osteoclastic bone resorption was not observed in any alveolar bone at any time.
