Protoplasts of Geotrichum candidum were obtained and cultivated in thin-layer agar following the method of Fukui et al. Electron microscopic observations were made on the regenerating protoplasts with the following results. 1. The protoplast was surrounded by a unit membrane, and contained mitochondria microsomes, vacuoles and a nucleus. 2. The regenerating cells which became osmoresistant during cultivation contained many distinct microsomes and mitochondria with well-developed cristae, and the cytoplasm was electron dense. These features indicate vigorous metabolism in the cells. 3. Regenerating cells which became osmoresistant after cultivation for 4-6 hr contained several vacuoles differing in electron density, mitochondria, microsomes, nuclei and cytoplasm that increased in electron density during maturation, while their daughter cells were still connected to the mother cells. Some of the regenerating cells had a cytoplasmic membrane of high electron density. 4. The osmoresistant cells were classified into two types on the basis of their ultrastructure. One type had a regular array of granules on the electron dense cytoplasmic membrane. The other had some electron dense material arranged irregularly on the cytoplasmic membrane. 5. Most of the regenerating cells which became osmoresistant after cultivation for 7 hr had a complete cell wall but it was only one third as thick as that of normal cells. This seems to indicatae that cell wall synthesis takes place within one hour.
