There is no doubt that chemotherapy for oral cancers is an important therapy following surgery and radiation. However, since there is to date no anti-cancer drug available, the effect of which is restricted to the cancer cells alone, there are a number of cases where the normal tissues and cells are also affected and even some cases lead to serious side effect. For the foregoing reason, the author cultivated first the skin tissue of fetus of albino rat by tissue culture method. Then various anti-cancer drugs were applied to the cultivated epithelium-like cells and fibroblast-like cells and the effect of the drugs on cell proliferation and morphology was investigated. Conclusions were reached as follows : 1. With regard to Nitromin (NMO), both the epithelium-like cells and fibroblast-like cells were restricted in proliferation in each experimental case, in both the high-concentration application group (1.0 mcg/ml, HA Group) and low-concentration application group (0.1 mcg/ml, LA Group), compared with the control group. The trend was greater in the HA group. Such regressive changes as swelling, deformity, pyknosis and vacuolation of the nucleus ; dispersion of the nucleolus ; vacuolation, disintegration of the cytoplasm and abnormal cell division were observed as the effects on the cells. These changes were greater in the HA group and in proportion to the number of application days. 2. With regard to Mitomycin (MT-C), restrictions in proliferation of both cells were observed in both the HA group (0.04 mcg/ml) and LA group (0.004 mcg/ml). In the LA group, however, findings suggestive of stimulatory proliferation were observed in 3-day application cases. An the effect on the cells, swelling, pyknosis deformity, vacuolation, disappearance of chromatin and agglutination of the nucleus ; dispersion or multiplicity of the nucleolus were significant. Regressive changes such as vacuolation and disintegration were observed in the cytoplasm. The changes were greater in epithelum-like cells in particular and in proportion to the number of application days in the HA group. 3. With regard to Bleomycin (BLM), restriction in proliferation in the HA group (0.3 mcg/ml) was obvious. In the LA group (0.03 mcg/ml), a trend for stimulatory proliferation was observed in 3-and 5-day application cases. In 7-day application cases, the fibroblast-like cells proliferated significantly compared with the epithelium-like cells, and the over-all proliferation was close to that of the control group. As the effects on the cells, regressive changes such as swelling, deformity, pyknosis and vacuolation of the nucleus ; dispersion of the nucleolus ; vacuolation the cytoplasm were observed in both cells. Observations of these changes were more obvious in the HA group and in proportion to the number of application days. 4. With regard to 5-Fluorouracil (5-Fu), such proliferation of cells as seen in the control group was not observed in HA group (5.0 mcg/ml) or in LA group (0.5 mcg/ml). Indication of stimulatory proliferation, which was observed in the cases of low-concentration application of NMO, MT-C and BLM, was not obseved at all. Regressive changes such swelling and pyknosis of the nucleus disappearance of chromatin ; dispersion of the nucleolus ; vacuolation of the cytoplasm were observed in greater intensity in any experimental case in proportion to the number of application days. 5. Dilantin sodium had an accelerating prolific effect on the cells in both the HA group (200 mcg/ml) and LA group (20 mcg/ml). The HA group in particular showed significant proliferation. In the LA group, the extent of proliferation was slightly above that of the control group but no significant trend was not observed. A greater prolific trend was observed in the fibroblast-like cells than in the epithelium-like cells. As to the changes in the cells, the findings were similar to those of the control group and no remarkable changes were observed.
