The follwings were the results of observation on the human labial gingiva of maxillary incisors or of cuspid of 62 cases in total, male 36, female 26, from 3 month to 82 year-old, which were divided by anatomical characteristic into three Groups : the Infant Group of pre and post eruption of deciduous teeth ; the Adult Group of full eruption of permanent teeth ; and the Aged Group of edentulous or with denture. Optical microscopic observation a) The keratinization tended to increase with aging, but on the contrary decreased in the Aged Group. High keratinization was observed in some of gingivae with denture. Morphological changes of the median and the basal layers of the gingival epithelium showed no differentiation with aging. b) The width of the epithelium became thin with aging, but in the Aged Group, it became thick. The A/B ratio, as shown in diagram 1, increased with aging, and decreased in the Aged Group. The width of the epithelium was in inverse proportion to the A/B ratio. c) The Infant and Aged Groups were found to have, on the average, approximately 50% more mitotic index than the Adult Group. d) Periodic acid-Schiff reaction revealed positive in the Infant and the Aged Groups but did not react in the Adult Group. e) Melanin pigmentation in the gingiva was observed in 13 cases of the Adult Group and only in one case in each of the other two Groups. f) A decrease of cellularity and an increase of fibrous component with aging in the lamina propria were observed. In the Aged Group infiltration of lymphocytes frequently appeared. Electron microscopic observation The fine structure of the basal layer to the superficial layer of human gingiva was observed and changes of aging were examined. a) In the Adult Group, there were most numerous tonofilaments, which formed into bundles called tonofibrils. In the Infant Group, there were a few tonofilaments. In the Aged Group, the frequency was between the other two Groups. b) Intercellular space was the widest in the Adult Group. Differentiation on intercellular space was not evident between the Infant and the Adult Group. c) In the Adult Group, Intercellular junctions with desmosomes were numerous but comparatively fewer in the Infant and the Aged Groups. Consequently, in the latter two Groups, more microvillis than in the Adult Group were observed. d) At the desmosomes, it appeared that the fine filaments, which seem to be part of tonofilaments, entered the desmosomes and penetrated into adjacent cells. e) Melanocytes which involved premelanin in the basal cell layer were observed, and Langerhans cells which involved characteristic granules (Langerhans granules) in the median cell layer were also observed. But in the basal cell layer, Langerhans granules were also observed even in melanocytes.
