Several modified antimony trichloride reagents using ethylene chlorohydrine, α-dichlorohydrine, dichloroethylene, trichloroethylene, tetrachloroacetylene and chloroform as solvents and adding to these reagents 2% volume of acetyl chloride, are examined for the estimation of vitamin D by the spectrophotometric procedure. The results are as follows : (1) The modified antimony trichloride reagents using dichloroethylene, trichloroethylene and tetrachloroacetylene as solvents react stronger with vitamin D_2 and D_3 to give the same salmonpink color exhibiting an absorption maximum at 500 mμ as in the other reagents. The E^<1%>_<1cm> 500mμ values are the highest at the first 30th second after starting to add the reagents to the vitamin samples and are unstable within the first 1 minute, but then remaining constant for 2 to 5 minutes ; those values are twice as much as the value obtained by using the Nield's reagent. (2) The antimony trichloride-dichloroethylene reagent and the antimony trichloride-tetrachloroacetylene reagent react with cholesterol to give a yellowish orange color exhibiting absorption maximums strongly at 420 mμ and faintly at 500 mμ, but the antimony trichloride-trichloroethylene reagent and the Nield's reagent give an orange color exhibiting absorption maximums weakly at 420 mμ and strongly at 500 mμ. The former reagents react slowly with cholecterol to give low E^<1%>_<1cm> 500 mμ values and the latter reacts rapidly to give high E^<1%>_<1cm> 500mμ values. (3) The 25% antimony trichloride-dichloroethylene reagent is the most stable in these reagents when they are reserved at room temperature. This reagent added with absolute ethyl alcohol reacts strongly with vitamins D_2 and D_3 to give a higher initial optical density after approximately 30 seconds and the E^<1%>_<1cm> 500mμ value decreases rapidly. (4) It is possible to obtain satisfactory results in the spectrophotometric examination of low vitamin D potency oils using the 25% antimony trichloride-dichloroethylene reagent and the 25% antimony trichloride-tetrachloroacetylene reagent.
