It is commonly known that the diastatic decomposition of starch by mold is based upon the co-operative actions of α-anylase, β-amylase and maltase. But the mixing proportion of these partial enzymes will be varied by spcies and culture conditions.We investigated iodinc color change and saccharifying powr upon soluble starch, using enzyme solutions obtained by water extraction from wheat-bran cultures of several industrially important species of mold, namely, an yellow aspergillus (Asp. oryzae sp.) a black aspergillus (Asp. amamori sp.), an amylomyces (Rh. javanicus Takeda) and a white aspergillus (so-calbed albino variant of certain awamori balck species). Thus we obtained the following curves.From these curves, we can see, that the diastase of Asp. oryzae is composed chiefly of α-and β-amylases, those of black aspergillus and amylomyces chiefly of β-amylase and maltase, and white aspergilus contains a large quantity of the three partial enzymes.In addition we verrified Kitano and Tokuka's view, that Ohlsson's methods of separation of partial enzymes applied on malt-diastase, is not applicable on malt-diastase, because α-amylase of mold is less stable in heat than that of malt.
