An active water-insoluble saccharase-tannic acid complex was prepared by precipitating the enzymes, and a continuous inversion of sucrose was carried out by using a column packed with this insoluble saccharse-tannic acid comlex.It was revealed that the continuous enzyme reaction using a saccharase column was superior to the batch system using native saccharase.There was no significant difference between the native saccharase and the insoluble complex in regard to the optimum pH and temperature, but the complex was more active at the alkaline side and at a higher temperature, and less active in the acid region with a pH below 4.0 than the native saccharase. The native saccharase was fairly stable with a pH between 3.0 and 4.0,while the resistance of the complex against acid was extremely decreased and this resistance was dropped easily by keeping the pH below 4.0 and by dissociation and degradation of the enzyme molecule. It was also observed that the enzymatic resistance against pronase or urea was increased and that the insoluble complex was activated by a mild treatment with a mixture of urea and sucrose.The inhibitory action of fructose, glucose and mercuric chloride of the insoluble saccharase was lower than on the native saccharase.
