We studied copper uptake by the yeast Debaryomyces hansenii. Yeast cells were suspended in a solution of copper sulfate for a short period, collected on a membrane filter, washed with distilled water by filtration, and then incubated on the filter. After various indubation periods, the cells were washed with 0.15 M sodium chloride, and copper contents in the cells and the filtrate were measured. Copper content in the cells increased, while copper in the filtrate decreased, with incubation time. The result indicates the presence of two steps in copper uptake by the cell; rapid binding of copper to the cell, presumably to the cell surface, and slow transport of bound copper into the cell. The binding of copper to the cell surface was affected not only by the concentration of copper but also by the presence of amino acids, especially histidine and cysteine. On the other hand, the copper transport depended on temperature, pH (optimum at pH 5.0), the amount of copper bound to the cell surface, and cell age. The copper transport was inhibited by N-ethyl-maleimide, monoiodoacetate, and glutaraldehyde. This suggests the participation of carrier and a thiol compounds in this copper transport process.
