The degradative steps of ring fission of p-cresol and m-cresol by Trichosporon cutaneum WY 2-2 were examined.1)36 hr and 50 hr were needed, respectively, to reach maximum growth of this strain on media containing p- and m-cresols, and it took longer times compared with a medium containing phenol.The results show that the toxicity of cresols against WY 2-2 is more severe than that of phenol, and the adaptability of this strain towards a p-isomer is superior to that towards a m-isomer.2)During the growth of WY 2-2 on media containing p- or m-cresols, 4-methylcatechol, the hydroxylation product of p- or m-cresols, was detected in the growth media.This suggests that through the degradation of cresols by this strain, an additional hydroxyl group is introduced into the aromatic ring of cresols without oxidaton of the methyl group itself.3)An oxidation product of 4-methylcatechol by a crude extract of WY 2-2 was identified as 3-methylmuconic acid from the following data.(a)A thin-layer chromatogram of the ether extract of the reaction mixture showed an unsaturated compound that co-chromatographed with authentic 3-methylmuconic acid.(b)The UV absorption spectra of the oxidatioin product and authentic 3-methylmuconic acid were identical.(c)The mass spectrum of the dimethylester of the reaction product of 4-methylcatechol, showed an identical molecular ion peak at m/e 184 corresponding to C_9H&ly;12>O_4,3-methylmuconic acid dimethylester.4)All crude extracts from this organism grown on p-cresol, m-cresol or phenol possessed an intradiol oxygenase, catechol-1,2-oxygenase, which showed the same substrate specificity irrespective of the growth substrate used.From the above results, the early oxidation steps of cresols by Trichosphoron cutaneum WY 2-2 might be proposed to be as follows : m-cresol and p-cresol are hydroxylated first to 4-methylcatechol prior to ring fission.The ring fission of 4-methylcatechol is catalyzed by an intradiol oxygenase, catechol-1,2-oxygenase, giving 3-methylmuconic acid.
