The characteristics of endo-cellulase(EC 3.2.1.4), β-glucosidase (EC 3.2.1.21) and β-xylosidase (EC 3.2.1.37)from Clostridium thermocopriae sp. nov. JT3-3(IAM 13577) were studied. The molecular weight by gel filtration chromatography is 51,000 for endo-cellulase and 50,000 for β-glucosidase, and the molecular weight by SDS polyacrylamide elecrtophoresis is about 46,000 for both the enzymes. The optimal pH is 6.5 for endo-cellulase and β-glucosidase, 5.5 for β-xylosidase. The endo-cellulase could rapidly hydrolyze sodium car-boxymethyl cellulase(CMC) of more than 100,000 molecular weight, but slowly hydrolyzed the CMC of 30,000 molecular weight and hardly hydrolyzed the CMC of less than 22,000 molecular weight. The K_m value of β-glucosidase for hydrolysis of p-nitrophenyl-β-_D-glucoside was 2.3 mM in the range of 1-7 mM substrate and 5.3 mM in the range of 7-50 mM substrate. A high concentration of glucose inhibited β-glucosidase in a way resembling competitive inhibition. A high concentration of xylose inhigitie β-xylosidase.
