Spectral Properties of Cytochromes in Candida lipolytica Mitochondria

The cytochrome system of mitochondria prepared from Candida lipolytica has been studied with the difference spectra between two kinds of reduced state, produced by succinate respiration and by the addition of ditionite, and an oxidized state. The amounts of cytochromes a+a_3 and cytochromes c+c_1 in both reduced states were almost equivalent, but those of b-type cytochromes reduced by succinate were less than 50% of those reduced by dithionite.It was also shown by appropriate difference spectra that in the yeast mitochondria at least three different components of the b-type cytochromes were present : 1) a component that is reduced partially with succinate and completely with dithionite (a-peak at 565 nm), 2) a component whose reduction is induced in the presence of antimycin A (a-peak at 568 nm), and 3) component(s) reducible exclusively with added dithionite (a-peak at 561 to 563 nm). Judging from their functional behavior, the first and second components, whose absorption maxima are specific for this strain, seem to correspond to cytochromes b-562 and b-566 in mammalian mitochondria, respectively.
