In order to examine the antigenecity of collagen, rabbits were sensitized with the preparation of collagen which was obtained by treating the bovine Achilles tendon with dilute acetic acid and examined histochemically and electron-micro-scopically. Threee immunizing methods were used: intraperitoneal injection, intramuscular injection with adjuvant of Al(OH)_3 and combination of both intraperitoneal and intramuscular injections. After 4 weeks, the complement-fixing antibody was proved to be positive in sera of the combined immunizing group, and after 8 weeks in each of the intraperitoneal and intramuscular groups. Arthus phenomenon were proved positive in all immunized rabbits, and was observed that the anti-collagen serum inhibited the fiber reconstruction from the collagen solution. In the next place, two different antisera were obtained by means of immunizing the rabbits with collagen which was prepared from the bovine Achilles tendon and from the rat tail tendon, respectively, anti-bovine and anti-rat collagen rabbits sera. For the purpose to examine whether each collagen has an immunologic sepecificity, various examinations were carried out such as complement fixation test, fiber reconstruction from collagen solution, morphologic reaction produced by the injection of anti-sera into the various animals, Results were proved to be positive only when the homologous collagen solution was injected into the immunizing rabbits, or homologous antigen was used in immunological test, and mononuclear cell infiltration was observed when the anti-rat collagen serum was injected ntracutaneouly into the rats, In cases when the serum was given intravenously into the rats, not only hemorrhages of several organs, but also fibrinoid change were observed. Furthermore, intracutaneous injection of the anti-bovine collagen serum into the bovine induced the swelling of connective tissue with thrombus and slight neutrophilic infiltration. As a result of the absortion test, it was shown that antibody was absorbed by homologous collagen, but not absorbed by heterologous collagen. The inhibition of antisera on the fiber reconstruction was observed only when antisera was added to the homologous collagen solution. From the above-mentioned findings, it is supposed that collagen has an antigenicity, and a species specificity as an antigen.
