Series of experiments for separation of incomplete antibodies from sera of tuberculous patients and of guinea pigs immunized with tubercle bacilli were carried out by the freezing-thawing procedure and ultracentrifugation technique. Glass tubes (3×250 mm) filled with serum sample set in a rack at incline of 45°or 90° were placed in a cold room at -5℃, -15℃ or 40℃. After complete freezing, 1et sera thaw gradually at room temperature. Repeated the same procedure five or ten times. Then freezed glass tubes were cut equally with a glass-cutter in three pieces, i.e. upper, middle and lower part, each of them was put in a test tube, and the freezed sera were thawed gradually at room temperature. The thawed sera were tested by the Middlebrook-Dubos' passive hemagglutination, Sindo-Middlebrook's hemolytic test and Sindo-Utahashi's detection of incomplete antibodies using tuberculin preparations as the antigen. Although it was thought that densitometric patterns of paper-chromatograph and protein content of sera obtained from the upper part might indicate rather dilution of sera, sera obtained from the upper part, in general, reacted positively only in both the hemolytic test and the detection of incomplete antibodies. It was considered, therefore, that practical separation of incomplete antibodies can be accomplished by such freezing-thawing procedure. Further, similar results were obtained also by ultracentrifugation 40,000 rpm for 4 hours.
