Novel Culture Techniques for Bone Tissue Engineering

Repairing miscellaneous tissue defects is still a great challenge in clinical practice because suitable autologous tissue is only available to a quite limited extent while allografts are still under the shadow of suspectable of infections in addition to risky immunological problems. Nowadays combination of BMO with porous ceramics is a much promising substitute for bone tissue. Currently it is important and urgent to obtain enough bone formation in the composites so that they can be applied in clinical practice. In order to increase bone formation in composites of BMO and porous ceramics we performed this study by novel culture techniques. Different low pressures were applied to porous HA blocks loaded by BMO. After subculture HA/BMO were implanted into subcutaneous sites of syngeneic rats. After a certain time these implants were then harvested for biochemical analysis of alkaline phosphatase (ALP) activity, bone osteocalcin (OCN) content, and histological analysis. The potential bone formation enhancement effects of composites of BMO and _-TCP by perfusion culture container were also tested. After soaked in cell suspension, blocks were randomly transferred into perfusion container. The peristaltic pump was adjusted to deliver 2 ml/hr fresh medium to the perfusion container. For testing in vivo, after a subculture in perfusion container for 2 weeks, the composites were subcutaneously implanted into syngeneic rats. The ALP activity and OCN content, and a histological analysis of these composites were analyzed at different time points. The higher ALP activity, OCN content, and more bone formation were found in the 100mmHg pressure group and the perfusion group. These results suggest that application of proper low pressure to subculture of BMO cells in HA blocks is beneficial to bone formation, and the perfusion culture system is a valuable and convenient tool for applications in tissue engineering
