[SHORT REPORTS]Involvement of C Kinase in the Induction of c-fos Gene Expression via A Kinase by Adrenocorticotropin

We found that adrenocorticotropin (ACTH) transiently increases c-fos mRNA in rat adrenals in vivo. This was proved to be mediated by A kinase in primary cultures of rat adrenal cells. However, other reports have shown that ACTH increases c-fos mRNA via C kinase in the Y-1 cells of mice. To investigate whether C kinase is involved in this c-fos mRNA increase by ACTH in normal rat adrenal cells, primary cultures were treated for 45 min with ACTH or phorbol ester (PMA). Although ACTH increased c-fos mRNA to 3.5 times the basel level, PMA, either alone or in combination with ACTH, did not. However, pretreatment with PMA for 24 hr to block the C kinase activity resulted in significant decrease in both the basel and ACTH-stimulated c-fos mRNA levels. In contrast, β-actin mRNA levels were scarcely affected by any of the treatment. There results suggest that in normal rat adrenal cells C kinase plays a permissive role in the signaling pathway of c-fos induction by ACTH.
