STUDIES OF CATECHOLESTROGEN METABOLISM DURING NORMAL PREGNANCY : CHANGES OF PLASMA CATECHOESTROGEN LEVELS AFTER DHA-S OR E_1-S INJECTION

37-39週の正常妊婦にestrone-sulfate(E_1-S)40mg(9例)およびdehydroepiandrosterone-sulfate(DHA-S)を一回静注し,血中estradiol(E_2),estrone(E_1),2-methoxyestradiol(2-mE_2),2-methoxyestrone(2-mE_1)の継時的動態を検討した.E_1-S負荷後血中E_1の増加が著明で30分で前値の1294±264%(SE)に達したが,E_2の増加は緩徐で60分で197±20%にとどまつた.2-mE_1は15分で有意の増加を示し,60分で1205±214%に達した.2-mE_2の1増加は緩徐で60分で268±53%にとどまつた.DHA-S負荷後では血中E_2の増加が著明で,15分で457±131%に達したが,E_1の増加は緩徐で120分で367±95%に達した.2-mE_1の増加は,E_1と類似の変動を示し,120分で有意の増加(345±140%)を示した.2-mE_2は120分まで有意の増加は認められなかつた.各estrogen分画間の相関性については,E_1-S DHA-S負荷群ともE_2-2-mE_2間に有意の差は認められず,E_1-2-mE_1,E_1-2-mE_2,2-mE_1-2-mE_2間には有意の相関性が認められた.DHA-S負荷後著明なE_2の増加にもかかわらず2-mE_1に比し,2-mE_2の有意の増加が認められないことからE_2はE_1に転換し,2-hydroxy estrogenに代謝されることが示唆された.又,各estrogen分画の相関性の検討から,2-hydroxy estrogenの生合成はE_2→E_1→(2OHE_1)→2-mE_1が主経路であることが強く示唆された.
